Experimental principle:
Filter a certain amount of sample with a filter membrane to intercept algae, grind and crush algae cells, extract chlorophyll with acetone solution, centrifuge and separate, and measure the absorbance of the extraction solution at wavelengths of 750nm, 664nm, 647nm, and 630nm. Calculate the concentration of chlorophyll a in water according to the formula.

Application of JXFSTPRP-YLS standard method for extracting chlorophyll:

1. Filtration: Add magnesium carbonate suspension before filtration to prevent pigment decomposition. Negative pressure cannot exceed 50kPa.
2. Extraction of chlorophyll: Place the filter membrane in the JXFSTPRP-YLS extractor, add 10ml of 90% acetone solution, and grind at 1500-2000rpm for 1 minute. Crushing phytoplankton and algae cells on the filter membrane. After grinding, soak and extract in a dark place at 4 ℃ for 2 to 24 hours. Final centrifugation clarification: Centrifuge 1000g for 20 minutes or filter, and transfer the supernatant to a stoppered glass vial.
